What is Kisqali? Kisqali (ribociclib) is a CDK4/6 inhibitor by Novartis for HR+/HER2-negative advanced or metastatic breast cancer. It is the only CDK4/6 inhibitor with demonstrated overall survival benefit in multiple subgroups.

📦 Available in 200 mg tablets. Standard dose is 600 mg (3 × 200 mg) once daily.

FDA-Approved Uses

  • HR+/HER2-negative advanced or metastatic breast cancer — with letrozole or anastrozole as initial endocrine-based therapy
  • HR+/HER2-negative advanced or metastatic breast cancer — with fulvestrant
Biomarker Note: HR+ (ER+/PR+) and HER2-negative status required. Only CDK4/6 inhibitor with demonstrated OS benefit in pre- and postmenopausal women.

How Does Kisqali Work?

Ribociclib selectively inhibits CDK4 and CDK6, halting cancer cell progression from G1 to S phase. Used on a 21/7 intermittent schedule. Demonstrated overall survival benefit in the MONALEESA trial program.

Dosage & Administration

600 mg orally once daily for 21 days, then 7 days off. Dose reductions (400 mg or 200 mg) for QTc prolongation or toxicity. Avoid strong CYP3A4 inhibitors.
